An in vitro selection of tolerant cal to the saline stress, at Solanum tuberosum L. cv. Kondor and cv. Spunta, were undertaken. For that, three types of explants resulting from vitroplants of the two varieties (sheets, stems, roots) were used. The medium of Murashige and Skoog, was appointed like basic medium.The 6-benzylaminopurine (BAP) (0,5 mg.l -1) and acid,-dichlorophenoxyacetic (2,4-D) (0,5 to 4 mg.l -1) were added to this basic medium to study the callogénèse and regeneration on the cal.
At the two varieties, the Bap/2,4-D combinations significantly affected the callogénèse.While being based on these first results, it is the combination 0,5mg.l -1 BAP/0,5mg.l-1 2,4-D which was most reactive, and they is the cal of stems and sheets which gave the best results concerning the regeneration of seedling. The selection the variable ones was carried out on the cal resulting from fragments of stems and sheets of the two varieties on medium ms added with the combination 0,5 mg.l -1 BAP/0,5 mg.l -1 2,4-D, and of different NaCl concentrations (0 to 18 g.l -1).Tolerant cal of stems was obtained on the medium containing 7g.l -1 of NaCl for the Kondor variety, and on the medium with 9g.l -1 of NaCl for the Spunta variety.The tolerance of the selected cal was maintained after several subcultures.
