This present work aims to highlight the phytochemical compounds and the in vitro estimation of the anti-hemolytic activity of the extracts of the aerial part of a medicinal plant: Spinacia Oleracea L against hemolysis induced by hypotonia , H2O2 and heat. Phytochemistry results show that spinach is rich in tannins, flavonoids, terpenoids, alkaloids and sterols. The saponins were revealed at the level of the chloroform decoction, for the quinones they are present at the level of the aqueous decoction the anti-hemolytic activity against H2O2 is positive for all the dilutions of the aqueous decoction with a maximum at the dilution of 25%, the protection rate of which is 27%. The infusion extract diluted to 75% gave good protection of erythrocytes against hypotonic hemolysis (induced by NaCl) with a protection rate of 32.86%. The results of the anti-haemolytic activity in the presence of spinach extracts against heat indicate that whatever the dilutions for all the extracts, at a temperature of 60 C° the anti-haemolysis activity is weak. Components of Spinacia Oleracea extracts could be the main anti-hemolytic constituent. Indeed, the determination of polyphenolic compounds by colorimetric method showed that the aerial part of Spinacia Oleracea is very rich in polyphenols and flavonoids. The latter are closely related to the anti-hemolytic activity of spinach expressed by plant extractsen_US
